Human acellular amniotic membrane incorporating exosomes from adipose-derived mesenchymal stem cells promotes diabetic wound healing

Abstract Background Diabetic wounds threaten the health and quality of life patients their treatment remains challenging. ADSC-derived exosomes have shown encouraging results in enhancing diabetic wound healing. However, how to use effectively is a problem that needs be addressed at present. Methods A mouse skin model was established. (ADSC-Exos) were isolated, vitro application evaluated using human umbilical vein endothelial cells (HUVECs) dermal fibroblasts (HDFs). After preparation characterization scaffold acellular amniotic membrane (hAAM) loaded with ADSC-Exos vitro, they transplanted into vivo healing phenomena observed by histological immunohistochemical analyses identify mechanism exosome-hAAM composites. Results The hAAM dressing very suitable for delivery exosomes. enhanced proliferation migration HDFs promoted tube formation HUVECs vitro. In from showed hAAM-Exos accelerated regulating inflammation, stimulating vascularization, promoting production extracellular matrix. Conclusion Exosome-incorporated scaffolds great potential healing, while also providing strong evidence future clinical applications
